As a singer Thérèse Van Den Staepele - Van Mechelen studied with Amélie Thérèse Martin, the mother of the composer Émile Mathieu. Later she moved to Paris, like so many others in those days. There she perfected her singing technique with the legendary tenor Gilbert Duprez, who since 1855 directed a singing school of his own in Paris. She is said to also have had contacts with Gioacchino Rossini.

Van den Staepele composed the opera Le mauvais oeuil (The Bad Eye), which has remained unperformed, and a number of songs. The song collection Joies et tristesses (Joys and Sorrows), six charming 'mélodies' on texts by the French writer Armand Silvestre, was published by F. de Aynssa in Brussels. The library of the Antwerp Conservatory has preserved two copies, one of which contains a hand-written dedication to the composer and musicologist Léon de Burbure dated 16 November 1882.
